Quantitative fluorescence spectroscopy of living bacteria by optical condensation with a bubble-mimetic solid–liquid interface

Microbial interactions are closely related to human health, and secreted signal molecules from bacteria determine the gene expression of following bacterial cell density molecule density. However, conventional quantitative analysis number requires several days using standard cultivation methods, detection via microbial is difficult since they in extremely small amounts. In this study, we performed local fluorescence spectroscopy quantitatively evaluate assembly dispersoids (fluorescent microparticles bacteria) under optical condensation at a solid–liquid interface on our developed bubble-mimetic substrate, which exhibits low thermal damage after few minutes laser irradiation. The obtained results showed that intensity spectrum was positively correlated with concentration even when only tens assembled were observed. Furthermore, calibration curve by plotting integrated integrating over observed wavelength, living analyzed. clarified mechanism will pave way for rapid precise their biomolecules labeled fluorescent dye.
